Why on earth do that? What do you gain by using tight slots on all the domino's especially if you are marking by hand. You've only got to be slightly out and the joint won't go together which you usually find out when everything is already covered in glue. The dominos in this instance are purely to help align the boards and so I would only do the first slot tight and all the subsequent ones on both boards on the second setting using the cross stops - no marking out. This is one of the advantages of the domino over a biscuit jointer - no time wasted setting out.
