Having made some cutting boards for home & gift purposes, the answer is:  It depends.  Structurally, I don't think that Domino's add any value.  But I used the Dominos for alignment purposes, both vertically & horizontally.  It that context, I found them very useful.

How did you ensure that the domino did not get exposed on the end grain? 
 
GK: I am not sure what you mean by your question, but I will answer what I think is your question. If I am off target, please let me know.

The Domino's are internal to the cutting board, several inches from the ends of the cutting boards.  They are not exposed.  As far as food safety issues go, they may as well not exist.  The food only "sees" maple & purple heart.
